Maddam Sir Drama series starts with five police officers, including four females and one male officer—S.H.O. Haseena Mallik, S.I. Karishma Singh, Sub Inspector Pushpa Singh, and Constable Cheeteshwar Chaturvedi—working at a Mahila Police Station in Lucknow. They are assisted by Billu Champat. Haseena Malik is emotive when solving problems, while Karishma Singh has a short temper, which can occasionally lead to conflict.
DSP Anubhav Singh, an IB secret agent, and Karishma’s cousin attempt to reconcile their disputes. They love each other sincerely despite their disagreements. Haseena quickly gains Karishma’s trust as a devoted friend who will always stand up for her and protect her from harm. The Thana keeps resolving different cases. Soon, Haseena Malik and Anubhav Singh fall in love. Anubhav accepts Haseena’s proposal, but she later finds out that he lied about being in love with her to serve a national security task. After completing the task successfully, Anubhav recognizes that he made a mistake by using Haseena for the task and that he had been in love with her all along.
But she declines the proposal. However, as time goes on, their relationship improves and they get closer. Finally, Haseena agrees to Anubhav’s proposal, and the two begin to make plans for their wedding. Anubhav is instructed to call off his wedding and depart his city for a duty of national defense on the day of their marriage. Being powerless, he abandons Haseena because staying with him could endanger her life too though, according to Karishma. And there is a pause in their story. But after a while, events pull them back together.

When Thana hires ASI Mira to see if teaching her about human feelings is effective, a new case arises. Karishma believes the robot is attempting to replace actual police personnel. Together, Karishma and Meera solve the case, with the latter applying Haseena’s ideology and handling the cases in her manner. As she makes her way back to the police station, Haseena runs across a secret agent who needs to give a hard drive containing information on every secret agent in the nation. The mastermind who ordered Mira to kill Haseena is the head of the criminals who assassinated the guy after failing to obtain the disc from him.
After a month, Pushpa and Karishma meet Urmila Mahadev in Mumbai. To help her find Haseena’s killers, Karishma comes up with a scheme to disguise Urmila as Haseena Malik and take her to Lucknow. Soon on, it is discovered that Urmila is Haseena. To apprehend the terrorists who also attempted to assassinate Haseena for the hard disk drive having important data on the agents that, in the hands of the wrong people, may pose a threat to national security, she masked herself on the advice of her superior, secret agent Ajay Bakshi. Bakshi believes that a member of her station is responsible for hacking Mira.

Haseena chases after them and finds Karishma wearing Chingari Gang’s saree whereas the cheetah does the same. Karishma is told to carry on the act by Haseena. Binni-Cheetah is fired from her position after Shivani finds out more about her relationship. Afterward, Binni decided to leave the Chingari Gang, but she informed Shivani about Karishma’s betrayal by contacting her.
After Karishma is saved by Haseena, Shivani attempts to kill Haseena but accidentally kills Saira Begum. Shivani chooses to accept full responsibility for the actions of the Chingari Gang and turns herself into the MPT.
